La ausencia de inhibidores de la PCR se verificó mediante la amplificación del gen endógeno 16S (Trejo-Saavedra, 2002) (Figura 2). Para el control positivo (C. nucifera) (y para las muestras problema), se amplificó el fragmento de 315 pb, e indicó que el ADN estaba libre de sustancias inhibidoras de la PCR (como etanol, fenoles, nucleasas y otros compuestos degradantes) (Ausubel et al., 1999). El control negativo no mostró amplificación. Detección de fitoplasmas Dado que no se observó amplificación por PCR directa (utilizando los dos cebadores P1/P7), se realizó PCR-nesting para todas las muestras analizadas (excepto para el control positivo). El ADN del control positivo se extrajo de plantas de coco infectadas y se caracterizó por unos títulos elevados de fitoplasmas. Además, no se observó amplificación en el control negativo.

Se observó un fragmento de 1200 pb en la PCR anidada para el control positivo y las muestras problema. Por lo tanto, se detectó fitoplasma en el 40% de las plantas analizadas (Figura 3). Los restos negativos podrían estar asociados a otros patógenos, como hongos, virus o bacterias no cultivables (como Candidatus Liberibacter solanacearum) (Secor et al., 2009).

They maintain that the knowledge acquired and the application of new techniques are improving the cultivation of crops on the plots, which benefits the poorest sectors.

To carry out these functions effectively, close coordination is maintained with federal and state agencies.    In addition, it develops regulations and carries out actions to ensure compliance with current regulations. Among these are the development of activities to oversee the methods of final disposal of used water and waste generated by industrial and agricultural activities.
